The construction of roads causes changes in the landscape of the ecosystem, in extreme cases it causes erosion and loss of productive capacity of the soil. For this type of project, the EEIAs recommend revegetating the right-of- way with native species as a mitigation measure. In the case of the construction of the Senkata and Mantecani road section, Baccharis incarum, among others, are identified for these purposes. In this sense, the objective of this work was to evaluate the in vitro propagation from germination, multiplication, rooting and acclimatization and to evaluate the propagation by seeds in substrates under nursery conditions. The seeds were sown in MS medium, for the multiplication the T1 (0.2 mg L-1 BAP) were used; T2 (0.2 mg L-1 BAP+0.01 mg L-1 ANA); T3 (0.4 mg L-1 BAP) and T4 (0.4 mg L-1 BAP+0.01 mg L-1 ANA). For rooting, T1 (75 % MS+30 g sucrose) were used; T2 (75 % MS+0.5 mg L-1 AIB+30 g sucrose); T3 (75 % MS+0.5 mg L-1 AIB+50 g sucrose) and T4 (75 % MS+0.5 mg L-1 AIB+70 g sucrose). During acclimatization, substrates T1 (3:2:1) and T2 (3:2:2) of black earth, sand and worm humus were used, respectively. For sowing in the nursery, the T1 substrate was used. Under in vitro conditions at 42 days it presented 48.47 % germination, T2, in the multiplication it obtained a greater number of shoots during the second subculture (9.4) with 35.8 mm of shoot length. T2 presented a greater number of roots (8.1) with less length (29.7 mm). The two substrates presented a 97 % survival. Under nursery conditions, 30 % emerged after 4 days of planting.
