Abstract

Abstract The papaya production area in the Alto Beni region has decreased thanks to the diseases that affected the area. Biotechnology accelerates conventional programs for mass plant propagation. One way to increase plant production in short times is through somatic embryogenesis. The objectives of the research were: 1) to evaluate different disinfection methods for the in vitro establishment of Red Lady and Salvietty papaya zygotic embryos. 2) To determine the in vitro behavior of the Red Lady and Salvietty varieties through the embryogenic development of the established explants. The research was carried out in the biotechnology laboratory of the Sapecho Experimental Station. The evaluated varieties were; Red Lady and Salvietty, the experimental design used for the contamination and survival variables was a completely randomized design (DCA) with a bi-factorial arrangement (3x2) generating six treatments, the simple DCA was also used for the embryogenic development variables where generated two treatments. The selected explants were subjected to three disinfection methods proposed by different authors. The 50% medium (MS) was used as the basal medium, to which 2,4-D, L-Glutamine, Mioinositol were added for the formation of calluses and somatic embryos (dark environment), and 6-BAP, Mioinositol, and Riboflavin photoperiod 16 light hours and eight dark hours. The disinfection method proposed by Garcia (2013) achieved the highest survival percentage, achieving 65% of established explants. Regarding development, the best result was presented with the Red Lady variety, forming six calluses and 43 somatic embryos per explant, 11 germinated somatic embryos per sample. It is concluded that; The in vitro behavior of this species through somatic embryogenesis is mainly influenced by the variety, and the stage of development of the explant.
