Abstract Tuberculosis is an infectious-contagious disease, caused by several species of the Mycobacterium tuberculosis complex, today it is estimated that one third of the world population representing a threatto public health, mainly by the emergence Multidrug-resistant strains (MDR-TB) that express resistance to Rifampicin and Isoniazid simultaneously. Al-though currently molecular methods are used for the detection of this infectious agent, in our medium there are no studies about the application of molecular methods for the accurate and effective detection of MDR-TB strains. The present study intends to standardize the Real Time PCR technique by Melting Curves Analysis for the detection of mutations in specific genes that cause resistance to these antituberculosis drugs. Eleven strains of Mycobaterium tuberculosis phenotypically identified by culture and primary controls of the method for the determination of resistance Isoniacida and Rifampicina by changes of temperature of dissociation were used. The results of this standardization showed that the method used can be op-timized in reagent performance and correlates more than 80% with phenotypic results. The standardized method improves the time detection of tuberculosis and allows to obtain resistance profiles to the main first-line antituberculosis drugs.
