Abstract This research work was carried out at the BIORENA Germplasm Bank Laboratories at the Center of Research and Innovation in Agricultural Sciences - Villa Carmen Yotala located at 15 km from the city of Sucre, from 2017 to 2019. The objective was to develop a methodology that allows maintaining and preserving potato explants (Solanum tuberosum L subsp. Andigend), under minimum growth conditions. The experimental design used in this reaseach was bifactorial with 10 potato accessions (12-A, Imilla negra, Bol 1207, Morena, Chiar Imilla, Yungay, Sani Imilla, 6-B, Vera, Imilla) and 3 concentrations of osmotic stabilizers in the middle of conservation (MS with Mannitol 40gr/l, MS Sorbitol 30gr/l and Sorbitol 40gr/l) with 4 repetitions. Within the research method, the sprouting of tubers was induced, which were sown with a pH of 5.7 and kept in the growth room for 30 days with physical photoperiod conditions of 16 light hours and 8 dark hours at 21 °C of temperature, light intensity of 3200 lux, and 50-70% relative humidity. Besides, six multiplications were made from the vitroplants obtained using cuttings smaller than 1 cm, kept in the growth room. Finally, the vitroplants were placed in the conservation médiums, using half nodes (medios nudos) for eight months in the growth chamber. It is concluded that the accessions showed desirable characters in the conservation médiums with Mannitol 40 gr/1 with an average height of 17,39 mm, 7 leaves per vitroplant, but fewer roots in the accessions (3,97 roots). The conservation médium with Sorbitol 40gr/l was the one that obtained the highest number of roots in the accessions with an average of 5,16 roots. It is concluded that the conservation médiums with satisfactory results for conservation by the minimum growth method for the 10 accessions of potatoes with which it was worked, was the conservation Médium with Sorbitol at 0,4.
